The Mechanism of Leukocytosis Induced by Prednisolone

Abstract
On the mechanism of leukocytosis there have been presented a few posturation, such as nervoushumoral regulation and stress theory, however it remains so far unsolved. Recent studies revealed that leucocytosis can be induced easily by 11-oxysteroid and these studies projected a new foot-lights into this particular field.
Shu and Takashi demonstrated the presence of leukocytosis-inducing factor in the blood after triamcinolone injection but did not elucidated the source of the substance. Author intended to clarify the mechanism of leukocytosis by 11-oxysteroid and prednisolone was used as 11-oxysteroid in this study.
Method and materials : Mongrel dogs weighing 4.9^13.2 kg. were used. The- determination of W. B. C.. count, R.B.C. count, hemoglobin, hematocrit and differential count of W. B. C. were performed respectively by Barker-Ziirk¢¥s , apparatus, Sahli method, Wintrobe method and Giemsa and Wright stainimg. The determination of nucleic acid of thoracic duct fluid and blood plasma were done by S-T-S method (Schmidt-Thanhauser-Schneider¢¥s combined method).
Result : Part I. The effect of prednisolone on the number of leukocytes was observed in normal dogs, in the dogs with thoracic duct fistula, in the splenectomized dogs with thoracic duct fistula and splenectomized dogs with Eck¢¥s shunt and thoracic duct fistula.
1. Prominent neutrophilic leukocytosis proceeded by lymphopenia by prednisolone was confirmed in normal dogs. However the leukocytosis by prednisolone was markedly inhibited in the dogs with thoracic duct fistula.
2. Splenectomy and Eck¢¥s shunt noticeably inhibited the leukocytosis inducing action of prednisolone in the dogs with thoracic duct fistula.
Part II. As it is well known fact that 11-oxysteroid induces the lymphocytolysis, author also intended to confirm the effect of prednisolone on lymphoid tissue. Histological study was done on the lymph node, thymus and spleen from the dogs treated with prednisolone.
Marked destruction of small lymphocytes and hemorrhage in lymph follicles of lymph node and in white pulp of spleen and in cortex of thymus were observed in these prednisolone treated dogs.
Part III. From above mentioned histological findings of lymphoid tissue, author assumed that nucleic acid and its split poducts might be released into thoracic duct fluid and circulating blood from the lymphoid tissue. On the other hand there are many reports concerning leukocytosis-inducing action of nucleic acid and its split substances such as adenosine, guanosine and adenylic acid. Therefore author intended to confirm whether nucleic acid is increased in the thoracic duct fluid and blood plasma of the dogs treated
with prednisolone.
1. In the normal dogs treated with prednisolone there was marked increase of RNA and slight increase of DNA in thoracic duct fluid and slight increase of RNA and DNA in blood plasma.
2. In the dogs with thoracic duct fistula, the splenectomized dogs with thoracic duct fistula, the splenectomized dogs with thoracic duct fistula and Eck¢¥s shunt prominent increase of nucleic acid was confirmed in the thoracic duct fluid while only slight increase of nucleic acid in the blood plasma was observed.
